Evaluation of the onset and duration of response to cold air inhalation challenge in Cynomolgus monkeys (Macaca fascicularis)

Description:A method was developed for performing cold air challenge in cynomolgus-monkeys for potential use in future toxicology studies of the effects of inhaled agents on pulmonary toxicity. The temporal pattern of cold air inhalation challenge (CAIC) pulmonary effects in monkeys was also studied and the correlation between cold air challenge pulmonary effects and methacholine reactivity were investigated. Pulmonary function testing was performed at 10, 25, 40, and 55 minutes post challenge. Significant increases in average pulmonary flow resistance and decreases in dynamic compliance were observed with maximum impairment occurring at 25 minutes post challenge, with a trend toward a return to baseline values at 55 minutes post challenge. The authors demonstrated that CAIC can be performed in monkeys using challenge methods similar to those used in humans. Statistically significant bronchoconstriction occurred from CAIC monkeys, and no refractory period for repeat challenge occurred.
