Effects of 2-methoxyethanol on fetal development, postnatal behavior, and embryonic intracellular pH of rats

Description:This study was designed to investigate the teratogenic and behavioral teratogenic effects of 2-methoxyethanol (109864) (2-ME) alone and in combination with ethanol (64175) when administered in liquid diet as well as to determine 2-ME effects on embryonic intracellular pH. Female Sprague-Dawley-rats were exposed to 2-ME in liquid diets on gestation days seven through 18. Visceral or skeletal malformations were investigated in day 20 fetuses. Total embryomortality was produced by exposure to concentrations above was 0.006 percent or 16mg/kg. Additional rats were given 0.006 or figure eight activity and the Cincinnati water maze on postnatal days 48 through 65. About a 50 percent mortality was noted at the higher dose level; the dose level also increased the number of errors in the maze. No other behavioral effects were noted at either dose level. Efforts were made to determine whether simultaneous exposure to 2-ME and ethanol would reduce the teratogenicity of 2-ME. No reduction was noted. On injecting 0.33 milliliters/kilogram of 2-ME into rats on gestation day 13, and determining embryonic intracellular pH at 2, 4, 8, and 24 hours thereafter, an increase was noted in pH at 4 hours, but not at later time points. When amiloride was administered to block the sodium/hydrogen antiporter, 2-ME induced twice the incidence of cardiovascular malformations in fetuses than 2-ME alone. At doses which were nonteratogenic, developmental toxicity was noted to persist, but only limited behavioral teratogenicity was observed.
