Description:An analysis was made of the effects of benzamide (55210) (BAM), sodium-butyrate (156547) and dimethyl-sulfoxide (67685) (DMSO) on chondrogenesis in embryonic chick limb bud cells in culture using a computer imaging system. A micromass culture technique was used to prepare the cultures from stage 23/24 White-Leghorn-chicken embryos. There was a direct relationship between the degree of chondrogenesis and the amount of Alcian-blue staining. Low magnification images of individual micromass colonies were converted into derived images based on their percentage transmittance relative to the background. The derived images were analyzed for relative degree of chondrogenesis using as descriptors the area and integrated optical density (IOD). IOD was the preferred descriptor due to its accuracy over a wide range of threshold values. BAM markedly enhanced chondrogenesis in culture. Sodium-butyrate markedly inhibited chondrogenesis and DMSO produced mixed results. The authors suggest that the same descriptors may be used in quantifying data from various objects which can be stained in a selected and quantitative fashion. [Description provided by NIOSH]
