Antigen-specific stimulation of histamine releasing factors in diisocyanate-induced occupational asthma

Description:A study was conducted on the production of antigen specific mononuclear cell derived histamine releasing factor (HRF) following exposure to diisocyanates. HRF production was assessed in blood cells obtained from methylene-diphenyldiisocyanate (MDI) exposed workers with confirmed occupational asthma (OA), those with unconfirmed OA, and asymptomatic workers. No significant differences were seen between subject groups in the HRF activity in peripheral blood monocyte culture supernatants. Antigen specific stimulation of HRF production was seen in seven of eight subjects with confirmed OA compared with no subjects in the other exposed groups. Specific immunoglobulin-E and immunoglobulin-G (IgG) antibodies were seen in two of nine asymptomatic and four of 11 symptomatic workers and one of nine asymptomatic and eight of 11 symptomatic workers, respectively. Associations were demonstrated between the presence of antigen specific HRF activity enhancement and confirmed OA as well as between the presence of specific IgG antibodies against diisocyanates and a positive history of OA. An analysis examining cellular sources of diisocyanate stimulated HRF indicated that lymphocytes and adherent cells were the major sources of spontaneous and antigen stimulated HRF. The authors conclude that antigen specific HRF is a useful biomarker for occupational asthma induced by exposure to diisocyanates. [Description provided by NIOSH]
