Description:A conceptual framework for the validation and use of biologic markers was delineated with emphasis on the concept of a continuum of events between ambient exposure to a xenobiotic substance and resultant health impairments. Topics in this review included: research strategies for the use of biologic markers in toxicologic and epidemiologic research; reliability of biologic markers; validation of biologic markers; use of multiple markers; susceptibility of components in the continuum; epidemiological designs (longitudinal cohort studies, case/control studies, cross sectional studies, and hybrid studies); use of a meta analysis; and analytical issues. The author concludes that biologic markers are useful in etiologic and mechanistic research, in secondary prevention of disease, in risk assessment, and in assessing the effectiveness of environmental controls. The next stage in the development of biologic markers can be stimulated by a strategy for validating markers and the development of a framework for their application. [Description provided by NIOSH]
