Effects of a water extract of cotton bract (CBE) in isolated guinea-pig trachealis smooth muscle (GPT).

  Guinea-pig trachealis smooth muscle was used to compare the effect of ashed or dialyzed cotton bract extract (CBE) on the contractile response with the response induced by crude CBE. CBE effects on purinergic receptor mediated muscle relaxation was also examined. The smooth muscle contraction induced by the dialyzable extract was generally similar to that recorded for the crude extract, except for a 1mg/ml dose of the former which resulted in significantly less contraction than the crude extract. Nondialyzable and ashed extracts were of lower potency than the other two forms of extract, with the ashed extract being three fold less active than crude extract in terms of median effective concentration. The untreated extract potentiated the smooth muscle relaxation induced by adenosine-triphosphate and adenosine, but the same extract failed to potentiate the action of isoproterenol on the smooth muscle preparation. In fact, the presence of the extract resulted in a reduction in the sensitivity of the preparation to isoproterenol. The authors conclude that the CBE contains products which may interact as agonists with postjunctional purinergic receptors. [Description provided by NIOSH]
