Description:The effects of chromium (7440473) and manganese (7439965) particles on the interferon system were studied in mammalian cells. Rhesus- monkey kidney cell monolayers were pretreated with 0 to 2.0 milligrams per milliliter (mg/ml) chromium or manganese particles. They were then inoculated with influenza virus and assayed for interferon induction. Influenza virus growth and interferon production were studied in kidney cell monolayers that had been pretreated for 24 hours with chromium or manganese particles. Kidney cell monolayers were pretreated with 100 units of neuraminidase to remove sialic-acid residues and then treated with chromium or manganese particles and a viral inducer. Interferon production was measured. Chromium or manganese particles were mixed and incubated with sialic-acid. The particles were then sedimented, removed, and used to treat kidney cell monolayers. The monolayers were assayed for interferon activity. Chromium particles at a concentration of 0.5mg/ml and manganese particles at 0.1mg/ml inhibited interferon induction by approximately 50 percent. The rates of virus multiplication in metal treated and control cultures were similar, reaching plateaus after 20 to 24 hours. The final extent of viral growth attained in cultures treated with manganese or chromium was almost twice that of control cell cultures. Neuraminidase treatment of cell monolayers and sialic-acid pretreatment of chromium and manganese particles abolished the adverse effects of the metal particles on viral interferon induction. The authors conclude that chromium and manganese particles adversely affect the production of viral induced interferon. The receptivity and interaction of cell membrane bound sialic-acid residues with metal particles are involved in the cellular protective responses to chromium and manganese particles.
