Description:The effects of three natural products from cotton on the functional properties of alveolar macrophages were investigated. The effects of gossypol (303457), rutin (153184), and catechin (1208090 were studied. Alveolar macrophages from male Sprague-Dawley-rats were incubated with and without cotton dust, aqueous extracts of cotton dust, or chemicals for 15 minutes at 37 degrees-C. Zymosan was added to the mixture before chemiluminescence measurements were taken to determine the release of reactive compounds. Oxygen consumption and mean cell volume were measured. Activity of acid- phosphatase, beta-glucuronidase, and beta-N-acetyl-glucosaminidase was monitored. Rate of phagocytosis was measured. Cotton dust and extracts assumed large amounts of oxygen even in the absence of alveolar macrophages. Exposure of cells to 0.0001 moles catechin, rutin, and gossypol had no effect on cellular volume. Catechin nor rutin had an effect on oxygen consumption of resting or zymosan stimulated macrophages. Particle stimulated cells were more sensitive to gossypol than the other cotton products. Only acid- phosphatase activity was inhibited by gossypol and rutin. Catechin was ineffective. All three cotton products were potent inhibitors of chemiluminescence. This inhibition was dose dependent. Gossypol had a greater inhibitory action than rutin. Catechin had the least inhibitory action. All products inhibited phagocytosis. The authors conclude that significant impairment of macrophage function can occur after exposure to two of the three natural products. [Description provided by NIOSH]
