Improved resolution in high performance liquid chromatography analysis of polynuclear aromatic hydrocarbons using ternary solvent systems.

Description:The resolution of the high performance liquid chromatography (HPLC) technique employed during the analysis of polynuclear aromatic hydrocarbons (PAHs) was improved via the optimization of solvent selectivity. PAHs, being structurally similar compounds produced by most incomplete combustion processes, have been identified out as potent carcinogens. The necessity of simultaneously detecting numerous PAHs at extremely low concentrations has been cited as one of the most challenging tasks standing before the occupational health chemist. In this text, the ternary acetonitrile/methanol/water solvent system employed during PAH analysis was examined over a wide range of concentrations in order to determine an optimum mobile phase composition. Paired fluoranthene/pyrene, benz(a)anthracene/chrysene, and perylene/benzo(a)pyrene compounds were used to put together a standard solution containing roughly 0.1 milligram per milliliter of each PAH. An isocratic ternary acetonitrile/methanol/water solvent system was found which afforded maximum resolution of the fluoranthene/pyrene peaks at a capacity factor of 5. Two additional isocratic ternary solvent systems were found which optimized the resolution of each of the other two PAH compound pairs. A solvent gradient was constructed that incorporated all three isocratic compositions, these compositions being determined simultaneously by means of a simple statistical design strategy. The initial mobile phase composition of the solvent system constructed was 48.4 percent acetonitrile, 7.8 percent methanol, and 43.8 percent water, with a final mobile phase composition of 43 percent acetonitrile, 37 percent methanol, and 20 percent water. The author concludes that selection of a ternary solvent system for the HPLC analysis of PAHs within these limits will result in improved HPLC resolution values. [Description provided by NIOSH]
