Description:The relationship between red blood cell membrane concentrations of lead (7439921) and activities of sodium-potassium-adenosinetriphosphatase (Na+,K+-ATPase) was investigated. Erythrocytes were prepared from blood samples taken from 8 normal human subjects, 14 lead exposed workers without lead poisoning, 5 exposed workers with lead poisoning and high blood lead concentrations, and 5 exposed workers with lead poisoning and low blood lead concentrations. Lead concentrations in various erythrocytic fractions were determined, and membrane Na+,K+-ATPase activity was measured. An inverse correlation was found between membrane lead concentrations and Na+,K+-ATPase activity, but no relationship existed between enzyme activity and total erythrocytic lead. The residual lead in the red blood cells was evenly divided between a high molecular weight protein and the membrane. The authors suggest that the primary determinant of lead toxicity in a given cell is the distribution of lead among nontoxic binding proteins and compartments that contain susceptible enzyme systems.
