Sister chromatid exchange in murine alveolar macrophages, regenerating liver and bone marrow cells - a simultaneous multicellular in vivo assay.

  A method for simultaneously affecting sister chromatid exchange (SCE) in murine alveolar macrophages, regenerating liver and bone marrow cells of hepatectomized mice and in alveolar macrophages and bone marrow cells of nonhepatectomized mice was developed. Both nonhepatectomized and hepatectomized BDF1-mice were given 9 hourly, 2 milliliter injection of bromodeoxyuridine (59143) (BrdU). The following day, the animals were injected with 100 micrograms of colchicine (64868) and sacrificed. Liver, lung and bone marrow cells were isolated. Differential labeling of sister chromatids occurred simultaneously in alveolar macrophages regenerating liver and bone marrow cells of partially hepatectomized mice and in alveolar macrophages and bone marrow cells of nonhepatectomized mice. The percentage of second division cells depended on cell type, and no significant differences were apparent between hepatectomized and nonhepatectomized mice. BrdU incorporation did not significantly alter the mitotic yield or relative cell density of macrophage preparations. The authors note that this multicellular assay is useful in simultaneously assessing the relative mutagen induced effects in a variety of tissues. [Description provided by NIOSH]
