Description:The mutagenic effects of dimethylnitrosamine (62759) (DMN), 2- aminoanthracene (613138) (2-AA), and 7,12-dimethylbenz(a)anthracene (57976) (7,12-DMBA) on cultured Chinese hamster lung cells were investigated at the hypoxanthine guanine phosphoribosyl transferase (HGPRT) locus. All three compounds induce 6-thioguanine (6TG) resistance only in the presence of rat liver microsomes. The mutation frequencies induced by the compounds are dose dependent. The authors conclude that DMN, 2-AA, and 7,12-DMBA require metabolic activation for mutagenicity. They suggest that the mutagenic potential of carcinogens and other chemical agents can be detected and quantitated through the use of Chinese hamster cells and microsome activation.
