Description:The activation of the oxidative burst of polymorphonuclear granulocytes (PMN) by supernatants of quartz (14808607) or coal mine dust exposed cultures of human mononuclear cells were investigated. The morphological changes of PMN under influence of quartz dust induced mononuclear cell supernatants with a transmission electron microscope were also evaluated. Quartz dust DQ 12 was used as fibrogenic stimulus for cultures of human mononuclear cells. The results indicated that human monocytes/macrophage in culture release a soluble mediator following incubation with quartz dust DQ 12 or coal mine dust TF-1. The mediator(s) stimulates human PMN to the release of reactive oxygen species, especially of superoxide anion. This mediator was identified as granulocyte activating mediator (GRAM). The relative thermoresistance of GRAM and the sensitivity of GRAM against treatment with protease or chymotrypsin suggested a protein nature of GRAM. Besides the enhancement of the oxidative burst of PMN by GRAM, the induction of strong chemotactic changes was also noted along with the release of lysosomal products form GRAM treated PMN in a time dependent manner by ultrastructural analysis. The authors state that these results present an in-vitro example of possible nondirect mechanisms of quartz and coal mine dust pathogenicity.
